Solid question...requires a top five. In no particular order:

When Harry Met Sally
Love Actually
Notting Hill (and I hate Julia Roberts, so that's saying something)
While You Were Sleeping (yes, really)
Keeping the Faith (very very good call, Brett)

Honorable mention to anything starring Katherine Heigl and/or Ryan Reynolds. If Dane Cook is in it, even better. Yes, the previous two statements were sarcastic.

I was going to be assy and write "Sophie's Choice" which then made me think about Kevin Kline and how fantastic he is. In and Out is a great romantic comedy.

Then that leads to Selleck and thus Three Men and a.... I also loved Joe vs. the Volcano as a kid.

More recently, I enjoyed Friends with Benefits.
