I really liked the Christina Applegate show Samantha Who? I thought that ended too soon. And of course Arrested Development. Luckily, Christina and Will Arnett will be in some show on ABC in the fall.
I would say Friday Night Lights but I think they are winding down to a nice conclusion. High school dramas typically have a built in expiration date. 90210 is the exception that proves the rule.
If I could have any show back that was not necessarily canceled it would be Lost (Season 1-3 style) or The Wire.

I think I am the only person who wanted to see more of Studio 60 on the Sunset Strip. Arrested Development would be up there too.
Quantum Leap.
Sports Night, definitely
Bug Juice: Our Summer at Camp starring Jonathan Adler
Oh, I forgot Studio 60. Good call, Matt. I really wish that show stayed on the air longer.
And Veronica Mars. Season one was the best, but I'd like to see what they came up with when she moved to DC.
DEADWOOD.
Or, Manimal (the man who can become any animal!)
seconding both sportsnight and freaks and geeks!
